We aimed to determine the developmental profiles of these PG synthases, their regulation by inflammatory mediators, and the PG synthetic capacity of rat serosal mesothelium in vitro. By real-time reverse transcription-polymerase chain reaction (RT-PCR), COX1 and COX2 messenger RNAs (mRNAs) were detected in fetal rats at gestational day (GD) 17 and day of birth (D0), i.e., prior to birth, in response to the proinflammatory mediators tumor necrosis factor-alpha, interleukin (IL)-1beta, and lipopolysaccharide, and in cultured rat serosal mesothelium. Prostaglandin (PG) synthesis, measured as formation of an immunoactive PG metabolite after [3H]arachidonic acid (AA) incubation, was higher at D0 than at GD17 in fetal rats, indicating that, from GD17 onward, a PG synthase other than COX1 is functionally expressed in rat serosal mesothelium. Tumor necrosis factor-alpha, IL-1beta, or lipopolysaccharide increased PG synthesis from GD17 to D0, with a more pronounced effect on PG synthesis at D0 than on PG synthesis at GD17. Morphology-independent PG synthesis was detectable in cultured rat serosal mesothelium. Both COX1 and COX2 mRNAs were detected in cultured rat serosal mesothelium
