There are several pathways that regulate autophagy and the genes associated with these pathways are often altered in cancer, including loss of tumor suppressor genes, inactivation of oncogenes and activation of oncogenes. Autophagy has been shown to be associated with therapeutic resistance, the main reason for which is the high level of autophagy in cancer cells, which is why they have high survival capacity and resistance to chemo and radiotherapy. Targeting autophagy can sensitize cancer cells to chemotherapy and radiotherapy. However, there are still many challenges, such as lack of understanding of the molecular mechanisms of autophagy in cancer and differences in genetic and phenotypic alterations between the many cancer types.
