It has long been recognized that a combination of in vitro and in vivo animal testing can provide valuable information on the safety and effectiveness of chemical compounds. Recent advances in chemistry and cell biology have enabled efforts to utilize novel types of cell-based assays in which specific human cell-types are utilized to assess the toxicological and pharmacological effects of compounds. In this review, we describe the current situation in cell-based assays for drug toxicity and efficacy and discuss the combination of cell-based and animal model approaches for pre-clinical drug testing.Dear Sir/Madam:

NIRF imaging was introduced in 2001 as a novel, noninvasive technology for optical visualization of biological structures and is based on low-light-level, near-infrared fluorescence from endogenous fluorophores (e.g., porphyrins, aminolevulinate acid (ALA), fluorochromes, flavins, and cytochrome c).

The initial applications of NIRF imaging have been in molecular/cellular biology to study cell-cell interactions and to monitor cancer cell growth, for example, to detect disseminated tumor cells (DTCs) in bone marrow in patients with solid tumors.^[@bibr1-1753425919864383]^

Until recently, the main role of NIRF imaging has been translational research in patients with malignancies, preclinical applications have been limited. For example, until recently, NIRF imaging was most useful for molecular imaging of bone marrow and sentinel lymph nodes in patients with melanoma.^[@bibr1-1753425919864383]^ Furthermore, there has been interest in the detection of premetastatic niche formation in non-melanoma cancers, in which metastasis occurs at a rate of 10% by 5 years in patients with breast cancer and the detection of metastatic niche cells may be used to prevent and treat cancer.
