Antigen binding to B cell receptors (BCRs) causes receptor aggregation and leads to B cell activation through the generation of highly phosphorylated and ubiquitinated phospholipase C?2 (PLC?2). Ubiquitination is a post- translational modification that is important in protein sorting and degradation. Recently we demonstrated that a proline-directed Ser/Thr phosphatase, PPP4C, dephosphorylates PLC?2 at the phosphoserine-173 (Ser-173) and deubiquitinates and maintains PLC?2 at the plasma membrane. Loss of PPP4C results in serum-independent NF-kB activation and lymphadenopathy in the B-cell receptor?deficient mouse strain. The studies proposed herein are centered on the regulation of PLC?2 ubiquitination, a key event in lymphocyte activation. The first aim will determine the molecular mechanism by which PPP4C regulates PLC?2. Preliminary data indicate that PPP4C binds to PLC?2 directly and that the activity of PPP4C is regulated by dephosphorylation at Ser-173. The second aim will identify the site on PLC?2 that is responsible for deubiquitination and is a regulator of PLC?2 activity. Preliminary data indicate that PPP4C deubiquitinates the PLC?2 transmembrane domain. The third aim will determine the role of PPP4C in T-cell activation. Preliminary data indicate that the TCR mediates NF-kB activation in a PLC?2-dependent manner and that PPP4C regulates TCR-dependent NF-kB signaling. The proposed work will provide novel insights into
