The activating mutation BRAF V600E is the most frequent genetic alteration observed in human cancers, including melanoma and thyroid cancers. Although the oncogenic potential of mutant BRAF has been attributed to the hyperactivation of the MAPK pathway, the molecular mechanisms by which mutant BRAF promotes tumorigenesis remain largely unexplored. Here, we demonstrate that oncogenic BRAF V600E mutant is sufficient to drive the formation of transformed cells in a 3-dimensional tissue culture environment. We show that the BRAF V600E mutant is present in both premalignant and malignant lesions in patients with neurofibromatosis type 1 (NF-1), a tumor predisposing genetic disorder. Furthermore, we show that the BRAF V600E mutation is present in benign neural neoplasms in NF-1 patients, suggesting that it is not a sufficient condition for malignant transformation. In contrast, benign neural neoplasms in NF-1 patients with a BRAF V600E mutation develop into malignant melanomas over time. Importantly, we demonstrate that the malignant transformation of cells expressing mutant BRAF V600E is abrogated when the mutant form of BRAF is not expressed. These findings provide a mechanism by which mutant BRAF can act as a tumor promoter in NF-1-deficient cells, and highlight the importance of elucidating the molecular mechanisms underlying the transformed phenotype.Local Blogs
